目的通过观测卡铂经腹腔给药后,对雌性大鼠子宫及血清中FSH、E2水平及卵巢形态的影响。方法选取3月龄雌性大鼠40只,随机分为A、B、C、D 4组,A、B、C为实验组,D为对照组;每日观察明确性周期,用药结束后24h,处死大鼠;取静脉血测FSH、E2,切取卵巢和子宫称量湿重、做病理切片、记数各级卵泡。结果A、B、C 3组大鼠体重、卵巢湿重、子宫湿重与D组相比明显减轻(P<0.05);A组卵巢湿重与B、C组相比亦有减轻(P<0.05)。A、B、C 3组的总卵泡数、成熟卵泡数、初级卵泡数和生长卵泡数明显少于D组(P<0.05);B、C两组(药量40 mg/kg)的成熟卵泡数较A组(药量20 mg/kg)减少(P<0.05);用药10 d的C组与用药5 d的B组相比,生长卵泡数明显减少(P<0.05),初级卵泡数则相对增加(P>0.05);A、B、C 3组的FSH值与D组比较均明显升高(P<0.05);3组E2值与D组比较显著下降(P<0.05);B、C组FSH水平明显高于A组(P<0.05),E2值则明显低于A组(P<0.05);A组和B组各级数卵泡均减少,B组伴间质及颗粒细胞的崩解坏死;C组生长卵泡、成熟卵泡数均明显减少或无成熟卵泡发育;A、B、C 3组,子宫粘膜层不同程度的变薄,上皮以柱状上皮为主,固有层腺体减少,腺腔小,结缔组织增多。结论卡铂所致的卵巢、子宫毒性与用药剂量及时间有关,主要表现为大卵泡数减少,子宫粘膜层变薄。
OBJECTIVE: To observe the effects of carboplatin administered intraperitoneally on the levels of FSH and E2 in uterus and serum of female rats and ovarian morphology. Conclusions The ovarian and uterine toxicities induced by carboplatin are related to the dose and time of administration. The main manifestations are the reduction of the number of large follicles and the thinning of uterine mucosa.
