目的建立检测8种常见皮肤分枝杆菌的PCR-RFLP方法。方法①用通用引物对分枝杆菌的热休克蛋白hsp65基因序列进行PCR扩增:②用限制性内切酶BstEⅡ和HaeⅢ对扩增产物进行消化,根据所得片段确定各分枝杆菌特征性的酶切图谱,从而对分枝杆菌进行鉴别。结果 8种常见皮肤分枝杆菌感染标准株(鸟、胞内、堪萨斯、结核、瘰疬、海、偶发、龟分枝杆菌)的酶切图谱与文献报道一致。结论 PCR-RFLP方法可用以检测鉴定皮肤分枝杆菌。
Objective To establish a PCR-RFLP method for the detection of eight common dermatophytes. Methods ① The universal primers were used to amplify the hsp65 gene of mycobacterial heat shock protein. ② The digestion of the amplified product was carried out by using restriction endonucleases BstEⅡ and HaeⅢ. According to the obtained fragments, the characteristic enzymes of mycobacteria Cut the map to identify mycobacteria. Results The restriction maps of eight common Mycobacterium dermatitis strains (birds, intracellular, Kansas, tuberculosis, gall bladder, sea, sporadic, Mycobacterium cheloneri) were reported in the literature. Conclusion PCR-RFLP method can be used to detect and identify Mycobacterium.
