目的探索自体血浆添加量和庆大霉素对培养的细胞因子诱导的杀伤细胞(CIK细胞)免疫表型和活性的影响。方法采用流式细胞术分析自体血浆和庆大霉素对CIK细胞中不同亚型免疫细胞比例的影响,细胞计数、MTT法检测处理后的CIK细胞对HeLa细胞的细胞毒性差异。结果自体血浆实验组与对照组之间不存在显著性差异,而庆大霉素实验组与对照组相比,CD3+细胞、CD56+细胞、CD3+CD8+细胞和CD3+CD56+细胞的数量均下降,差异有统计学意义(P<0.05),下降幅度分别为1.4、1.7、1.4和2.5倍。进一步研究发现,庆大霉素实验组与对照组的CIK细胞对HeLa细胞的细胞毒性分别为43.0%和90.0%,两者存在显著性差异(P<0.05)。结论自体血浆对CIK细胞不同亚型比例无明显影响,庆大霉素可引起CD3+细胞、CD56+细胞、CD3+CD8+细胞和CD3+CD56+细胞减少,有一定细胞毒性。
Objective To explore the effects of autologous plasma volume and gentamicin on the immunophenotype and activity of cultured cytokine-induced killer cells (CIK cells). Methods Flow cytometry was used to analyze the effect of autologous plasma and gentamycin on the proportion of immune cells in different subtypes of CIK cells. The cytotoxicity of CIK cells to HeLa cells was detected by MTT assay. Results There was no significant difference between the experimental group and the control group, while the number of CD3 + cells, CD56 + cells, CD3 + CD8 + cells and CD3 + CD56 + cells in gentamicin group were significantly decreased compared with the control group Statistically significant (P <0.05), the decrease was 1.4,1.7,1.4 and 2.5 times. Further study found that the cytotoxicity of CIK cells in gentamicin experimental group and control group on HeLa cells were 43.0% and 90.0%, respectively, with significant difference (P <0.05). Conclusions Autologous plasma has no significant effect on the proportion of different subtypes of CIK cells. Gentamicin can reduce the number of CD3 + cells, CD56 + cells, CD3 + CD8 + cells and CD3 + CD56 + cells, and have certain cytotoxicity.
