目的 建立一种敏感、特异、简便快速的囊虫病免疫诊断方法。方法 以胶体金标记抗猪囊尾蚴抗原单克隆抗体 ,将另一抗猪囊尾蚴抗原单克隆抗体在硝酸醋酸混合纤维素膜上固定成一条线 ,制成免疫测试条。待测抗原与金标单克隆抗体结合后沿着硝酸醋酸混合纤维素膜移动 ,与膜上的特异性单克隆抗体结合形成肉眼可见的褐红色线。结果 当猪囊尾蚴囊液抗原量为 5 ng时 ,测试条仍呈阳性反应。检测脑囊虫病患者脑脊液 (CSF)的阳性率为 82 .6 0 % ,检测健康人 CSF的阴性符合率为 10 0 %。将本法检测 CSF结果与 EL ISA比较 ,两方法的敏感性和特异性相似。结论 本方法敏感、特异 ,操作简便快速 ,试剂稳定 ,重复性好 ,在囊虫病的诊断中具有很好的应用前景
Objective To establish a sensitive, specific, simple and rapid immunological diagnosis of cysticercosis. Methods Monoclonal antibody to anti-Cysticercus cellulosae was labeled with colloidal gold, and another anti-Cysticercus cellulosae antigen monoclonal antibody was immobilized on a nitrocellulose mixed cellulose membrane to form an immunoassay strip. The antigen to be tested binds to the gold labeled monoclonal antibody and then moves along the nitrocellulose mixed cellulose membrane and binds to the specific monoclonal antibody on the membrane to form a maroon-colored line that can be visually observed. Results When the antigen of Cysticercus cellulosae was 5 ng, the test strip still showed a positive reaction. The positive rate of cerebrospinal fluid (CSF) in patients with cerebral cysticercosis was 82.60%, and the negative coincidence rate of CSF in healthy people was 100%. Sensitivity and specificity of the two methods are similar for comparing CSF results detected by this method with EL ISA. Conclusion The method is sensitive, specific, easy to operate, stable and reproducible, and has a good application prospect in the diagnosis of cysticercosis
