检测4种不同产地的山药——河南焦作的“铁棍山药”、山东菏泽的“陈集山药”、湖北武穴的“佛手山药”和江西瑞昌的“南阳山药”中尿囊素的含量。采用超声辅助提取,反相高效液相色谱法进行检测,色谱柱为PhenomenexLunaC18(250mm×4.6mm,5μm);流动相为甲醇-水(1∶9,V/V);流速0.5mL/min;柱温为室温;检测波长224nm。结果表明:尿囊素在1.596~7.182μg线性关系良好,R_2=0.9995;尿囊素的平均回收率为97.2%,RSD=1.20%(n=3)。该方法稳定性、精密度和加样回收率都良好,可以为山药质量控制提供可靠手段,各产地山药中尿囊素含量差异较大,其中南阳山药的尿囊素含量最高,比含量最低的铁棍山药高出124%。
