作者采用Westem-blotting方法,对一株抗人肺癌单克隆抗体WLA-2C4所识别的肿瘤相关抗原WLA-Agl进行了初步分析.用免疫组织化学方法对WLA-Agl在肺癌、肺外脏器肿瘤以及人体某些正常组织中的表达状态进行了研究.结果表明,WLA-2C4识别的抗原是分子量约58KD的糖蛋白。该抗原主要分布于肺低分化腺癌以及大细胞癌,在肺外脏器的某些低分化癌亦有较强的表达,而在肺内及肺外的高分化癌表达较少.在正常支气管、肺、胃肠粘膜等极少表达.提示由单克隆抗体WLA-2C4所识别的抗原WLA-Agl可能是一种新的肿瘤相关抗原。
The authors used Westem-blotting method to analyze the tumor-associated antigen WLA-Agl recognized by human anti-human lung cancer monoclonal antibody WLA-2C4. Immunohistochemistry was used to detect WLA-Agl in lung cancer and extra-pulmonary organ tumors. As well as the expression status of some normal human tissues, the results showed that the antigen recognized by WLA-2C4 is a glycoprotein with a molecular weight of about 58KD. The antigen is mainly distributed in poorly differentiated adenocarcinoma and large cell carcinoma of the lung. It is also strongly expressed in some poorly differentiated carcinomas of extra-pulmonary organs, but is less expressed in well-differentiated carcinomas in and outside the lungs. The bronchus, lungs, and gastrointestinal mucosa are rarely expressed, suggesting that the WLA-Ag1 antigen recognized by the monoclonal antibody WLA-2C4 may be a novel tumor-associated antigen.
