补中益气汤不同配伍对脾虚大鼠免疫器官及小肠黏膜转运体蛋白表达的影响

目的:探讨补中益气汤及其配伍对脾虚模型大鼠免疫器官及小肠黏膜中葡萄糖及肽转运体蛋白表达的影响。方法:建立脾虚大鼠模型,成模动物随机分模型组、补中益气汤组及补中益气汤去“升-柴”组和倍“升-柴”组,另设正常对照组,比较各组大鼠体质量及脏器指数,采用Western blot法检测大鼠小肠黏膜中钠依赖性葡萄糖糖转运体(SGLT1)、葡萄糖转运体2(GLUT2)、肽转运体1(Pep T1)的表达。结果:与正常对照组比较,模型组大鼠体质量及脾脏指数和胸腺指数均显著降低(P<0.05),补中益气汤各组大鼠脾脏指数显著升高(P<0.05);补中益气汤组和去“升-柴”组大鼠体质量及胸腺指数显著升高(P<0.05)。模型组大鼠小肠黏膜中SGLT1、GLUT2蛋白表达较正常对照组显著降低(P<0.05);与模型组比较,补中益气汤组SGLT1蛋白表达显著升高(P<0.05),补中益气汤各组GLUT2蛋白水平显著升高(P<0.05),各组Pep T1蛋白表达水平差异无统计学意义(P>0.05)。结论:补中益气汤及其配伍对脾虚大鼠体质量及免疫器官脏器指数降低和葡萄糖转运蛋白表达低下的异常改变有较好的改善作用,补中益气汤原方的效果优于其配伍方。 Objective: To investigate the effect of Buzhong Yiqi Decoction and its compatibility on the protein expression of glucose and peptide transporter in immune organs and intestinal mucosa in rats with spleen deficiency syndrome. Methods: The model of spleen deficiency was established in rats. The model animals were randomly divided into model group, Buzhong Yiqi Decoction group and Buzhong Yiqi Decoction group to go to “Sheng-Chai” group and times “Sheng-Chai” group. Normal control group, the body weight and visceral index of rats in each group were compared. The expressions of sodium-dependent glucose transporter (SGLT1), glucose transporter 2 (GLUT2), peptide transporter 1 Pep T1) expression. Results: Compared with the normal control group, the body weight, spleen index and thymus index in the model group were significantly decreased (P <0.05), while those in the rats in the Buzhong Yiqi Decoction group were significantly increased (P <0.05) The body weight and thymus index of Zhongyiqi Decoction group and the rats going to “Shengchai” group were significantly increased (P <0.05). The expression of SGLT1 and GLUT2 protein in intestinal mucosa in model group was significantly lower than that in normal control group (P <0.05). Compared with model group, the expression of SGLT1 protein in Buzhong Yiqi Decoction group was significantly increased (P <0.05) The levels of GLUT2 in each group were significantly increased (P <0.05), but there was no significant difference in the expression levels of Pep T1 between groups (P> 0.05). Conclusion: Buzhong Yiqi Decoction and its compatibility have a good effect on the improvement of body weight, immune organ index and abnormal glucose transporter protein expression in rats with deficiency of spleen, and the effect of Buzhong Yiqi Decoction is better than that of compatibility square.
