目的探讨石见穿醇提取物体外对血管内皮细胞生物学行为的影响。方法采用人脐静脉内皮细胞(HUVECs)原代培养模型,通过MTT法观察石见穿醇提取物对HUVECs及人肝癌HepG2细胞增殖的影响;通过Transwell小室迁移、体外小管形成实验,观察石见穿醇提取物对HUVECs迁移、小管形成能力的影响。结果 12.5-200μg/ml的石见穿醇提取物作用于HUVECs48h,细胞增殖抑制率为17.73%-59.70%,明显高于其作用于人肝癌HepG2细胞的7.19%-39.66%(P<0.01);12.5-200μg/ml石见穿醇提取物作用HUVECs 12h后,迁移抑制率为1.83%-64.51%,且抑制作用与药物浓度呈正相关(r=0.940,P<0.05);12.5-200μg/ml石见穿醇提取物作用于HUVECs 24h后,形成管腔数目明显减少,管腔不完整甚至消失。结论石见穿醇提取物在体外能够明显抑制HUVECs增殖、迁移和小管形成,改变血管内皮细胞生物学行为。
OBJECTIVE To investigate the effect of alcoholic extract of stone seeker on the biological behavior of vascular endothelial cells in vitro. Methods The primary cultured human umbilical vein endothelial cells (HUVECs) were cultured and the effects of Atractylodes macrocephalae extract on the proliferation of HUVECs and HepG2 cells were observed by MTT assay. Transwell chamber migration and in vitro tubular formation were observed to observe Effects of Alcohol Extracts on HUVECs Migration and Tubule Formation Ability. Results The inhibitory rates of 12.5-200 μg / ml Shijingshan alcohol extract on HUVECs 48h were 17.73% -59.70%, significantly higher than 7.19% -39.66% (P <0.01) of HepG2 cells. The inhibitory rate of HUVECs transfected with alcohol extracts from 12.5 to 200μg / ml for 12h was 1.83% -64.51%, and the inhibitory effect was positively correlated with drug concentration (r = 0.940, P <0.05); 12.5-200μg / ml stone See through the alcohol extract on HUVECs 24h, the formation of a significant reduction in the number of lumens, lumen is not complete or even disappear. CONCLUSION Ethanol extract can significantly inhibit the proliferation, migration and tubule formation of HUVECs in vitro and change the biological behavior of vascular endothelial cells.
