本文对23例恶性滋养细胞和19例正常早孕绒毛滋养细胞进行了原代培养。分别采用免疫组化、反转录PCR、放免法和酶联免疫吸附法,检测了体外培养滋养细胞P53蛋白和β-hCGmRNA的表达、β-hCG、hPL和IL-6含量的变化。体外培养的恶性滋养细胞中,细胞滋养细胞增生异常活跃,细胞核呈明显异型。恶性滋养细胞P53蛋白阳性表达,且阳性强度与正常早孕绒毛滋养细胞比较均有显著差异(P<0.01)。体外分泌β-hCG和IL-6的含量均显著高于正常滋养细胞(P<0.05)。P53蛋白的表达以及IL-6的分泌可能与恶性滋养细胞的异常增殖和低分化有关。
In this paper, 23 cases of malignant trophoblast cells and 19 cases of normal early pregnancy villous trophoblast cells were primary cultured. The expression of P53 protein and β-hCG mRNA, β-hCG, hPL and IL-6 in cultured trophoblast cells were detected by immunohistochemistry, reverse transcription PCR, radioimmunoassay and enzyme-linked immunosorbent assay respectively. Malignant trophoblast cells cultured in vitro, cytotrophoblast hyperplasia is extremely active, the nucleus was significantly atypia. The positive expression of P53 protein in malignant trophoblast cells was significantly different from that of normal trophoblast cells in early pregnancy (P <0.01). The levels of β-hCG and IL-6 secreted in vitro were significantly higher than those of normal trophoblast (P <0.05). P53 protein expression and secretion of IL-6 may be associated with abnormal proliferation and poorly differentiated malignant trophoblast cells.
