目的:研究白术对脾虚动物利水作用,探讨其可能作用机制。方法:采用优化后饮食不节结合劳倦过度法,造成动物脾虚模型。腹腔水负荷生理盐水建立脾虚水湿内停模型。肛温、体重、表观指标为脾虚模型大鼠的造模期评价指标。尿中D-木糖排出量、粪便含水率、血清总蛋白(TP)、白蛋白(ALB)、总胆固醇(TC)、高密度脂蛋白(HDL-C)、低密度脂蛋白(LDL-C)、水通道蛋白1、胃泌素(Gas)及肾髓水通道蛋白1(AQP1)等为研究指标,探讨白术调节脾虚大鼠胃肠功能的有效剂量及相关机制。结果:造模期,大鼠体重下降,肛温无变化,表明此方法造成动物脾虚。给药后,白术(3.6 g·kg-1)能略增模型大鼠尿量和尿中D-木糖排出量,明显升高粪便含水量、TP、ALB、HDL-C,但降低TC和LDL-C含量,略降低水通道蛋白1水平。腹腔水负荷试验,模型组动物,体重下降率显著降低,给予白术水煎液后则下降率显著升高,尿量略升高。结论:中剂量组白术水煎液有运化水湿作用,可有效改善动物模型脾虚状态,提高其相应消化吸收功能。初步认为白术排水途径可能由粪便排出。对大鼠进行腹腔水负荷后,说明白术有改善机体水液代谢能力,能有效改善TP、ALB、HDL-C、TC、LDL-C等指标。白术对水的调节作用不通过AQP1实现,且白术高剂量效果不及中剂量。
Objective: To study the effect of Atractylodes on the water of animals with spleen deficiency and to explore the possible mechanism. Results: During the modeling period, the body weight of rats decreased and the rectal temperature did not change, indicating that this method caused spleen deficiency in animals. After administration, Atractylodes macrocephala (3.6 g · kg -1) slightly increased urine volume and urine D-xylose excretion, increased stool water content, TP, ALB and HDL-C, but decreased TC and LDL-C content, slightly lower aquaporin 1 levels. Abdominal water load test, the model group of animals, weight loss rate was significantly reduced, given Atractylodes Decoction declining rate was significantly increased, slightly increased urine output. CONCLUSION: The middle-dose Atractylodes macrocephala decoction has the function of moistening and hydration, which can effectively improve the state of spleen deficiency in animal model and improve the corresponding digestion and absorption function. Initially that Atractylodes drainage pathway may be excreted. After peritoneal water load in rats, Atractylodes macrocephala has the ability to improve water and liquid metabolism, and can effectively improve the indexes of TP, ALB, HDL-C, TC and LDL-C. Atractylodes regulate the water does not pass AQP1 achieved, and the high dose of Atractylodes less effective than the dose.
