目的:研究重楼提取物对前列腺癌细胞DU145的增殖抑制作用及其可能机制。方法:采用MTT法检测重楼不同提取物对DU145细胞增殖的影响,同时运用克隆形成抑制实验观察正丁醇提取物对DU145细胞的增殖抑制作用,荧光倒置显微镜下观察加药前后细胞形态的变化,运用RT-PCR检测重楼正丁醇提取物对DU145细胞中Caspase-3、Survivin、BAX、BCL-2 mRNA表达的影响。结果:MTT结果显示,重楼不同提取物对DU145细胞的增殖均有一定的抑制作用,其中以正丁醇提取物的抑制作用最强;RT-PCR结果显示,重楼正丁醇提取物能促进DU145细胞Caspase-3、BAX mRNA表达并抑制Survivin、BCL-2 mRNA表达。结论:重楼提取物抑制DU145细胞增殖作用的机制可能与其促进Caspase-3、BAX mRNA表达及抑制Survivin、BCL-2 mRNA表达有关。
Objective: To study the inhibitory effect of extract of Rhizoma Paridis on proliferation of prostate cancer cell line DU145 and its possible mechanism. Methods: MTT assay was used to detect the proliferation of DU145 cells. The inhibitory effect of n-butanol extract on the proliferation of DU145 cells was also observed by clonogenic assay. The morphological changes of DU145 cells were observed under a fluorescence inverted microscope RT-PCR was used to detect the effects of n-butanol extracts from Rehmannia glutinosa on expression of Caspase-3, Survivin, BAX and BCL-2 mRNA in DU145 cells. Results: The results of MTT showed that different extracts of Rhizoma Paridis inhibited the proliferation of DU145 cells in a certain extent, of which n-butanol extract had the strongest inhibitory effect. The results of RT-PCR showed that n-butanol extract of Rhizoma Paridis Promote DU145 cells Caspase-3, BAX mRNA expression and inhibit Survivin, BCL-2 mRNA expression. Conclusion: The mechanism of the inhibitory effect of extract of Rhizoma Paridis on the proliferation of DU145 cells may be related to the promotion of the expressions of Caspase-3, BAX mRNA and the expression of Survivin and BCL-2 mRNA.
