目的:观察比较广枣中三种不同工艺提取物的抗心律失常作用,为找到一种抗心律失常作用较强的主要化学单体或简单复合物打下基础。方法:(1)乌头碱导致大鼠心律失常;(2)哇巴因导致豚鼠心律失常;(3)结扎冠脉导致大鼠心律失常。结果:样品1、样品2可显著延长乌头碱所致大鼠早搏、室速、室颤、心脏停搏出现时间,使哇巴因所致豚鼠心律失常出现时间延长,明显减少结扎大鼠冠状动脉所导致的室性异位搏动数、推迟心律失常出现时间、缩短室速时程、降低室颤发生率,并呈剂量依赖性。样品3使乌头碱所致大鼠室速、室颤出现时间缩短,使哇巴因所致豚鼠心律失常出现时间缩短,对结扎大鼠冠状动脉所导致的大鼠室性异位搏动数、心律失常出现时间和室速时程均有不同程度的负性影响。结论:样品1、样品2能对抗乌头碱、哇巴因、结扎冠状动脉等所导致的心律失常的发生;而样品3不仅没有对抗作用,还有可能促进心律失常的发生。
OBJECTIVE: To observe the antiarrhythmic effects of extracts from three different techniques of Polygonum orientalis and to lay a foundation for finding a major chemical monomer or simple compound with strong antiarrhythmic effect. Methods: (1) Aconitine induced arrhythmia in rats; (2) ouabain caused arrhythmia in guinea pigs; (3) Arrhythmia was induced in rats by ligation of coronary artery. RESULTS: Samples 1 and 2 significantly prolonged the occurrence of premature beats, ventricular tachycardia, ventricular fibrillation and cardiac arrest in rats induced by aconitine, and prolonged the occurrence of arrhythmias in guinea pigs caused by ouabain, which significantly reduced the coronary artery in rats. The number of ventricular ectopic beats caused by arteries, delaying the occurrence of arrhythmia, shortening the ventricular tachycardia duration, and reducing the incidence of ventricular fibrillation were dose-dependent. Sample 3 shortened the ventricular tachycardia and ventricular fibrillation induced by aconitine, shortened the arrhythmia time in guinea pigs caused by ouabain, and induced ventricular ectopic beats in rats induced by ligation of rat coronary arteries. The time of arrhythmia and the duration of ventricular tachycardia have different negative effects. Conclusions: Samples 1 and 2 can resist the occurrence of arrhythmias caused by aconitine, ouabain, and ligation of coronary arteries. Sample 3 not only has no antagonistic effect, but also may promote arrhythmia.
