目的研究无氧再灌对海马脑片的影响以及鞘氨醇和佛波酯的作用。方法采用海马脑片体外无氧再灌模型,以孵育上清液乳酸脱氢酶漏出量为损伤指标。结果体外培养脑片先无氧15min,再灌0~120min,随着无氧再灌时间的延长,LDH漏出增加。无氧前15min,于培养液中分别加入鞘氨醇和佛波酯,则LDH漏出量明显低于对照组。结论无氧和再灌引起海马脑片严重损伤,鞘氨醇和佛波酯对上述损伤有明显的保护作用
Objective To study the effects of anaerobic reperfusion on hippocampal slices and the effects of sphingosine and phorbol ester. Methods Anaerobic reperfusion model of hippocampal slices was used in vitro. The amount of leakage of lactate dehydrogenase in the supernatant was taken as the injury index. Results In vitro, brain slices were anaerobically exposed to oxygen for 15 min and then reperfused for 0 to 120 min. With prolonged anaerobic reperfusion, LDH leakage increased. 15min before anaerobic, sphingosine and phorbol ester were added to the culture medium, then the LDH leakage was significantly lower than the control group. Conclusion Anoxic and reperfusion induced severe damage of hippocampal slices, and sphingosine and phorbol ester have obvious protective effect on the above injury
