目的 研究宽筋藤提取物对牛Ⅱ型胶原诱导的类风湿性关节炎(CIA)模型大鼠的改善作用。方法 取大鼠随机分成5组,每组10只,即空白组,模型组,甲氨蝶呤组(1 mg·kg-1),宽筋藤高、低剂量组(宽筋藤提取物12,6 g·kg-1)。除空白组外,其余各组大鼠右后足底皮下注射牛Ⅱ型胶原与弗氏完全佐剂的混悬剂0.1 mL,7 d后在大鼠尾根部及背部分3点皮内注射等量的混悬剂。测量大鼠右后足掌和踝关节周长,每周1次;步态仪采集大鼠步态信息。HE染色观察大鼠膝关节和踝关节的病理变化。结果 宽筋藤提取物可以明显改善CIA大鼠足掌及踝关节肿胀度,缓解疼痛;明显减轻滑膜增生,减少炎细胞浸润及血管翳的形成,并改善软骨病变损伤。结论 宽筋藤提取物对CIA大鼠有显著的改善作用,对抗类风湿性关节炎药物开发有一定的意义。
Objective To study the effects of extract of Radix Gramineae on bovine type Ⅱ collagen-induced rheumatoid arthritis (CIA) rats. Methods The rats were randomly divided into 5 groups with 10 rats in each group: blank group, model group, methotrexate group (1 mg · kg -1) 6 g · kg -1). Except for the blank group, the rats in the other groups were injected subcutaneously with 0.1 mL of suspension of bovine type Ⅱ collagen and Freund’s complete adjuvant in the right hind paw, and then injected intradermally Amount of suspension. The right hind paw and ankle joint circumference were measured once a week. The gait information was collected from the gait. HE staining was used to observe the pathological changes of the knee and ankle in rats. Results Radix Glabrae Radix extract could significantly improve the swelling of the foot and ankle in CIA rats and relieve the pain. It could significantly reduce the synovial hyperplasia, reduce the infiltration of inflammatory cells and the formation of pannus, and improve the cartilage lesions. Conclusions The extract of C. sinensis has a significant effect on improving CIA rats and has certain significance to the development of drug against rheumatoid arthritis.
