探讨睡眠剥夺引起的神经元凋亡与相关基因表达的变化。采用TUNEL染色观察了快眼动睡眠剥夺大鼠海马神经元形态学变化 ,应用原位杂交、Westernblot法检测了快眼动睡眠剥夺大鼠海马bcl 2 ,baxmRNA ,MAPKs表达的变化。结果表明 :快眼动睡眠剥夺大鼠海马CA1 ,CA3区神经元阳性凋亡细胞数明显增多 ,bcl 2 ,baxmRNA表达明显增强 ,ERK活性降低 ,JNK蛋白表达量较对照组明显增高。提示睡眠剥夺可引起大鼠海马神经元凋亡。与凋亡相关的bcl 2 ,baxmRNA基因表达及MAPKs活性的变化可能涉及神经元的凋亡机制
To investigate the changes of neuron apoptosis and related gene expression induced by sleep deprivation. The morphological changes of hippocampal neurons in rats after REM sleep deprivation were observed by TUNEL staining. The changes of bcl-2, bax mRNA and MAPKs expression in hippocampus of rats after fast-moving sleep deprivation were detected by in situ hybridization and Western blotting. The results showed that the number of apoptotic neurons in hippocampal CA1 and CA3 area increased significantly, the expression of bcl2 and bax mRNA increased, the activity of ERK decreased and the expression of JNK protein increased significantly. Tip sleep deprivation can cause apoptosis of rat hippocampal neurons. Apoptosis-related changes in bcl 2, bax mRNA expression and MAPKs activity may be involved in neuronal apoptosis
