目的:探讨3′-HVR探针及Southern杂交技术应用于实验小鼠遗传质量监测的可行性。方法:采用人源α-珠蛋白3′-HVR探针,应用ECL试剂盒,以HRP标记进行Southern杂交,以化学发光法进行检测,对5种近交系小鼠(BALB/c、DBA/2、T739、TA1、615)、1种封闭群小鼠(KM)及2种免疫缺陷小鼠(BALB/c-nu/nu及C.B-17SCID)进行DNA指纹分析。结果:其不同近交小鼠的DNA指纹图各异,而同种近交系的不同个体的DNA指纹图相同,封闭群KM小鼠不同个体之间的DNA指纹图存在一定差异。BALB/c-nu/nu小鼠、CB-17SCID小鼠不同个体之间的DNA指纹图相同,并且与BALB/c小鼠的一致。结论:该探针及其Southern杂交技术可用于实验小鼠的遗传质量监测。
Objective: To investigate the feasibility of using 3’-HVR probe and Southern hybridization in genetic quality monitoring of experimental mice. Methods: The human hybridization was performed by using the human α-globin 3’-HVR probe, ECL kit and Southern blotting with HRP, and the chemiluminescence method was used to detect the expression of these genes in BALB / c, DBA / 2, T739, TA1,615), one closed group mouse (KM) and two immunodeficient mice (BALB / c-nu / nu and C.B- 17SCID). Results: DNA fingerprints of different inbred mice were different, while DNA fingerprinting of different individuals in the same kind of inbred strains was the same. DNA fingerprinting among different individuals in the closed-group KM mice was different. The DNA fingerprints between BALB / c-nu / nu mice and CB-17SCID mice were identical and consistent with BALB / c mice. Conclusion: The probe and its Southern hybridization can be used to monitor the genetic quality of mice.
