以‘红玉’芒果采后96 h的果皮为材料,采用RT-PCR技术从芒果(Mangifera indica)果皮中克隆到ζ-胡萝卜素脱氢酶基因(ZDS),其cDNA全长为1 883 bp,具有1 710 bp的开放阅读框(ORF),编码569个氨基酸.对芒果ZDS蛋白的亲水/疏水性进行预测,发现芒果ZDS蛋白所含的氨基酸亲水/疏水性主要介于+2.5~-2.6之间.通过在线软件对其进行了定位预测及二级结构和三级结构进行了预测,发现ZDS定位在叶绿体中,ZDS基因编码的蛋白质有16个螺旋,13个折叠,通过系统进化树分析发现芒果中ZDS基因编码的蛋白与柚子、甜橙、蜜柑等植物的亲缘关系比较近.“,”Using the peel of ‘Hongyu’ mango after harvest 96 h as the experimental materials,and with the technology of RT-PCR was cloned from the pericarp of mango (Mangifera indica) to Zeta carotene desaturase gene (ZDS).We find the full length of cDNA was 1 883 bp,with a 1 710 bp open reading frame (ORF) encoding 569 amino acids.The secondary structure and the three level structure were predicted by online software.It was found that the protein encoded by the ZDS gene showed 16 helices,and the 13 fold.Using online software to predict the hydrophilicity/hydrophobicity (the negative value indicates the hydrophilicity,the positive value indicates hydrophobicity) and locate the protein of mango ZDS.The result showed that the hydrophilicity/hydrophobicity of the amino acid from mango ZDS protein mainly among +2.5~-2.6,and the location of genes in chloroplast.The phylogenetic tree analysis showed that zeta carotene desaturase (ZDS) gene encoding the protein of magon has a closer genetic relationship with grapefruit,orange,mandarin and other plants.
