目的:从真菌松萝中提取地衣酸并研究地衣酸对体外培养前列腺癌PC-3M细胞增殖的抑制效应。方法:用细胞培养、细胞生长抑制实验(MTT法)观察PC-3M细胞形态、细胞生长密度和检测细胞生长抑制率,用~3H-TdR掺入法测定细胞增殖过程中DNA的含量。结果:与对照组相比,实验组PC-3M细胞形态异常、细胞生长密度降低,细胞生长抑制率增强,~3H-TdR掺入量明显减少,并呈剂量依赖关系,与地衣酸浓度的相关系数分别为0.9799与-0.9525。结论:地衣酸对体外培养PC-3M细胞具有抑制效应。
OBJECTIVE: To study the inhibitory effect of lichen acid on the proliferation of prostate cancer PC-3M cells cultured in vitro. Methods: Cell morphology, cell density and cell growth inhibition rate of PC-3M cells were observed by cell culture and cell growth inhibition assay (MTT method), and DNA content in cell proliferation was measured by 3H-TdR incorporation assay. Results: Compared with the control group, the morphology of PC-3M cells in experimental group was abnormal, the cell density was decreased, the cell growth inhibition rate was increased, the amount of ~ 3H-TdR incorporation was significantly decreased and in a dose-dependent manner, The coefficients are 0.9799 and -0.9525, respectively. Conclusion: The lichen acid has an inhibitory effect on PC-3M cells cultured in vitro.
