从浙江省豌豆和蚕豆病株上获得两个病毒分离物,编号为P-935和B-934.人工摩擦接种6科19种植物,P-935能侵染4科13种植物,B-934能侵染4科14种植物,除菜豆外,它们在这些植物上的症状十分相似。P-935和B-934的钝化温度为50-55℃,稀释限点为10 ̄(-3)-10 ̄(-4),体外保毒期分别为5天和4天。两个分离物均能由桃蚜以非持久性方式传毒,用昆诺藜作繁殖寄主均提取到了大量病毒粒子,提纯病毒粒子球形,平均直径25nm。病毒外壳蛋白均由两种亚基组成,分子量分别为42.5kD和21.2kD,感病叶片组织中观察到了无定形的泡状内含体,在琼脂双扩散试验中,P-935和B-934均能与蚕豆萎蔫病毒(BBWV)抗血清(血清型Ⅱ)形成清晰沉淀线。根据以上特性,P-935和B-934均被鉴定为蚕豆萎蔫病毒,且都属血清型Ⅱ.初步调查表明杭州郊区豌豆病毒病均由BBWV引起,蚕豆病毒病主要有BBWV和黄瓜花叶病毒(CMV)。
Two virus isolates were obtained from pea and faba bean isolates in Zhejiang province, numbered P-935 and B-934. Nineteen plants of 6 families were inoculated by artificial rubbing, P-935 was able to infect 13 families of 4 families, and B-934 was able to infect 14 families of 4 families. Their symptoms on these plants were very similar except for the kidney bean. The passivation temperature of P-935 and B-934 was 50-55 ℃, and the dilution limit was 10-3 (-10) (-4). The in vitro protection period was 5 days and 4 days respectively. Both isolates could be poisoned by Myzus persicae in a non-persistent manner. A large number of virions were extracted from the host of Quinoa, and the virions were purified to spherical shape with an average diameter of 25 nm. The viral coat proteins consisted of two subunits with molecular weights of 42.5 kD and 21.2 kD, respectively. Amorphous vesicular inclusions were observed in susceptible leaf tissues. In the agar diffusion test, P-935 and B -934 all formed a clear precipitation line with the BBWV antiserum (serotype II). Based on the above characteristics, both P-935 and B-934 were identified as faba bean wilt virus and both belong to serotype II. Preliminary investigation showed that the suburban pea virus disease in Hangzhou was caused by BBWV. The main pathogen of broad bean virus was BBWV and cucumber mosaic virus (CMV).
