目的建立常温下快速溶解保健食品胶囊壳,高效液相色谱法测定其中非法添加的二甲双胍、苯乙双胍、吡格列酮和罗格列酮4种化学合成降糖药物的方法。方法在碱性条件下,胶囊壳在25℃~30℃下超声4 min~6 min,溶解后经过沉淀蛋白质,用高效液相色谱法进行检测。结果二甲双胍和苯乙双胍的检出限为0.1μg/ml,罗格列酮和吡格列酮的检出限为0.2μg/ml。二甲双胍、苯乙双胍、罗格列酮、吡格列酮4种降糖药的保留时间分别为4.907 min、6.519 min、13.512 min、14.965 min,回收率分别为62%~76%、98%~120%、58%~69%、30%~38%。样品的总分析时间不超过1 h。结论本方法快速准确,操作简单,可用于检测保健食品胶囊壳中上述合成降糖药物的测定。
OBJECTIVE To establish a capsule shell for rapid dissolving health food at room temperature and to determine the four kinds of chemical synthetic antihyperglycemic drugs illegally added with metformin, phenformin, pioglitazone and rosiglitazone by high performance liquid chromatography. Methods Under alkaline conditions, the capsule shell was sonicated at 25 ℃ ~ 30 ℃ for 4 min ~ 6 min. After dissolution, the protein was precipitated and detected by high performance liquid chromatography. Results The detection limit of metformin and phenformin was 0.1μg / ml, and the detection limit of rosiglitazone and pioglitazone was 0.2μg / ml. The retention times of metformin, phenformin, rosiglitazone and pioglitazone were 4.907 min, 6.519 min, 13.512 min, and 14.965 min, respectively. The recoveries were 62% -76%, 98% -120% 58% ~ 69%, 30% ~ 38%. Total sample analysis time does not exceed 1 h. Conclusion The method is rapid, accurate and simple to operate. It can be used for the determination of above synthetic hypoglycemic agents in capsule of health food.
