本实验用一定浓度的苦参碱作用K562细胞16h, 24h,48h,采用分子生物学Northern Blot和Dot Blot杂交技术分析c-myc,N-ras及P53 mRNA在K562细胞诱导分化过程中表达水平改变,以探讨其表达水平与细胞生物学特性的关系。结果显示苦参碱作用K562细胞24h时,N-ras基因mRNA表达活化;P53-基因表达增强:作用48h时,c-myc mRNA水平表达明显受抑。说明苦参碱在诱导K562细胞分化启动时,能明显改变早期相关癌基因的表达。提示c-myc,N-ras和P53等癌基因是一类重要的细胞增殖、分化调控因素。
In this experiment, K562 cells were treated with certain concentration of matrine for 16 h, 24 h, 48 h, and the expression of c-myc, N-ras and P53 mRNA in K562 cells was analyzed by Northern Blot and Dot Blot hybridization To explore the relationship between its expression level and cell biological characteristics. The results showed that matrine activated K562 cells 24h, N-ras mRNA expression was activated; P53-gene expression increased: 48h, c-myc mRNA expression was significantly inhibited. Description of matrine in the induction of K562 cell differentiation start, can significantly change the expression of early related oncogenes. It is suggested that oncogenes such as c-myc, N-ras and P53 are important regulatory factors of cell proliferation and differentiation.
