目的探讨基因组DNA甲基化状态与人小细胞肺癌(small cell lung cancer,SCLC)H446/CDDP细胞获得性多药耐药的关系。方法分别提取人SCLC多药耐药细胞H446/CDDP及其亲代细胞H446的基因组DNA,应用甲基化敏感性随机引物PCR(MS-AP-PCR)方法检测其甲基化状态。结果 H446/CDDP细胞经酶解后的扩增谱带较H446细胞明显增多,且荧光强度较强,表明其基因组DNA甲基化水平较亲代细胞明显增高。结论 H446/CDDP细胞多药耐药性的获得可能与其基因组DNA甲基化水平增高有关。
Objective To investigate the relationship between genomic DNA methylation status and acquired multidrug resistance in human small cell lung cancer (HCC) H446 / CDDP cells. Methods The genomic DNA of human multidrug-resistant human multidrug-resistant cell line H446 / CDDP and its parental cell line H446 were extracted and their methylation status was detected by methylation-sensitive random primer PCR (MS-AP-PCR). Results The amplified bands of H446 / CDDP after enzymolysis were significantly higher than those of H446 cells and the fluorescence intensity was stronger, indicating that the methylation level of H446 / CDDP cells was significantly higher than that of parental cells. Conclusion The multidrug resistance of H446 / CDDP cells may be related to the increase of methylation level of genomic DNA.
