目的探讨紧密连接蛋白Claudin-1、Claudin-4和Caspase-3在结直肠癌组织中的表达及临床意义。方法应用免疫组化SP法,检测60例结直肠癌和20例正常结直肠粘膜组织中紧密连接蛋白Claudin-1、Claudin-4和Caspase-3的表达水平。结果 Claudin-1、Claudin-4在结直肠癌组中阳性表达率分别为76.6%、85.0%,显著高于正常组的20.0%、30.0%(P<0.01);而Caspase-3在结直肠癌组中表达阳性率为21.7%,显著低于正常组的60.0%;有淋巴结转移组Claudin-1、Claudin-4的表达阳性率分别为50.0%、55.0%,明显高于无淋巴结转移组26.6%、30.0%(P<0.05);Caspase-3在有、无淋巴结转移组阳性率分别为10.0%、15.5%(P>0.05);TNM分期一、二组间Claudin-1、Claudin-4和Caspase-3蛋白表达阳性率差异有统计学意义(P<0.05),而其它组间阳性表达率差异无统计学意义。结论 Claudin-1、Claudin-4的高表达在结直肠癌的发生发展过程中可能起着促进作用,其表达与淋巴结转移、TNM分期有关,Caspase-3可作为结直肠癌预后标志物。
Objective To investigate the expression and clinical significance of Claudin-1, Claudin-4 and Caspase-3 in colorectal cancer. Methods The expression of Claudin-1, Claudin-4 and Caspase-3 in 60 cases of colorectal cancer and 20 cases of normal colorectal mucosa were detected by immunohistochemical SP method. Results The positive rates of Claudin-1 and Claudin-4 in colorectal cancer were 76.6% and 85.0%, respectively, which were significantly higher than those in the normal group (20.0% and 30.0%, P <0.01) The positive expression rate of Claudin-1 and Claudin-4 in group with lymph node metastasis was 50.0% and 55.0%, respectively, which was significantly higher than that in non-lymph node metastasis group (21.7% vs. 60.0% , 30.0%, respectively (P <0.05). The positive rate of Caspase-3 in group with or without lymph node metastasis was 10.0% and 15.5% -3 protein expression positive rate difference was statistically significant (P <0.05), while the other groups the positive expression rate difference was not statistically significant. Conclusions High expression of Claudin-1 and Claudin-4 may play a role in the development of colorectal cancer. The expression of Claudin-1 and Claudin-4 may be related to lymph node metastasis and TNM staging. Caspase-3 may be used as a prognostic marker for colorectal cancer.
