目的 :观察金属硫蛋白 (MT)对肝脏缺血再灌注损伤 (HIRI)大鼠体内酶活性的影响 ,旨在为临床应用奠定理论基础。方法 :建立HIRI动物模型 ,并按体重给予不同剂量的MT( 1、2、5mg·kg- 1) ;观察血清转氨酶、抗氧化酶活性及脂质过氧化物含量改变。结果 :与HIRI大鼠比较 ,MT补充组动物血清谷丙转氨酶 (GPT)、谷草转氨酶 (GOT)和乳酸脱氢酶 (LDH)活性明显降低 ;血清和肝组织中丙二醛 (MDA)含量下降 ;全血GSH -Px活性及血清SOD含量升高。三个剂量组中以 1mg·kg- 1MT补充组效果最佳。大鼠血清GPT、GOT、LDH活性分别降为 ( 2 55.50± 12 2 .99)U·L- 1、( 2 0 5.4 0± 76.4 8)U·L- 1、( 2 4 69.50± 90 2 .0 7)U·L- 1,与HIRI组相比 ,P <0 .0 1。结论 :再灌注前适量补充MT可通过抑制HIRI大鼠脂质过氧化、增强抗氧化酶活性 ,从而减轻肝脏缺血性氧化损伤 ,对肝脏缺血再灌注有保护作用
Objective: To observe the effect of metallothionein (MT) on the enzyme activity in the liver of rats with ischemia-reperfusion injury (HIRI), and to lay a theoretical foundation for its clinical application. Methods: The animal model of HIRI was established and different doses of MT (1, 2 and 5 mg · kg -1) were given according to body weight. The changes of serum aminotransferase, antioxidant enzyme activity and lipid peroxidation were observed. Results: Compared with HIRI rats, the activities of GPT, GOT and LDH in the MT-supplemented group were significantly lower than those in the HIRI group. The contents of malondialdehyde (MDA) in serum and liver tissue were decreased ; Whole blood GSH-Px activity and serum SOD increased. The three dose groups to 1mg · kg-1MT supplement group best. The activities of GPT, GOT and LDH in the serum of rats were reduced to (2.55 ± 0.502 ± 0.99) U · L-1, (2.50 ± 5.40 ± 76.48) U · L-1, 0 7) U · L-1, compared with the HIRI group, P <0. Conclusion: The proper amount of MT before reperfusion can inhibit hepatic ischemia-reperfusion injury by inhibiting lipid peroxidation and increasing antioxidant enzyme activity in HIRI rats
