目的探讨姜黄素对卵巢癌微血管内皮细胞体外生长及乙酰肝素酶表达的影响。方法体外培养卵巢癌微血管内皮细胞,分为正常对照组和姜黄素组(浓度分别为2、10、50、250μg/ml),观察时间为加入姜黄素共孵育后24、48、72h。MTT法检测细胞增殖抑制率,流式细胞仪检测细胞周期变化,免疫荧光法和RT-PCR法检测内皮细胞乙酰肝素酶表达,生化法检测细胞上清液丙二醛(MDA)含量及乳酸脱氢酶(LDH)活力。结果姜黄素组细胞生长抑制率增加,有显著的剂量依赖效应(P<0.01)。姜黄素组增殖指数显著下降(P<0.01),乙酰肝素酶mRNA水平和蛋白水平显著下降(P<0.01),细胞培养上清液MDA水平及LDH活力显著升高(P<0.01)。结论姜黄素(2~250μg/ml)能显著抑制人卵巢癌微血管内皮细胞增殖活性,降低乙酰肝素酶mRNA转录和蛋白表达。
Objective To investigate the effect of curcumin on the growth of hepatoma microvascular endothelial cells in vitro and the expression of heparanase. Methods Ovarian cancer microvascular endothelial cells were cultured in vitro and divided into normal control group and curcumin group (concentrations of 2, 10, 50, and 250 μg/ml, respectively). The observation time was 24, 48, and 72 hours after the addition of curcumin. The cell proliferation inhibition rate was measured by MTT assay, the cell cycle was detected by flow cytometry, the heparanase expression was detected by immunofluorescence and RT-PCR, and the malondialdehyde (MDA) content in cell supernatant was measured by biochemical method. Dehydrogenase (LDH) activity. Results The growth inhibitory rate of curcumin group increased, there was a significant dose-dependent effect (P <0.01). The proliferation index of curcumin group was significantly decreased (P<0.01), heparanase mRNA and protein levels were significantly decreased (P<0.01), and the cell culture supernatant MDA level and LDH activity were significantly increased (P<0.01). Conclusion Curcumin (2~250μg/ml) can significantly inhibit the proliferation of human ovarian cancer microvascular endothelial cells and reduce the transcription and protein expression of heparanase mRNA.
