目的:探讨E-cadherin(E-cd)、PTEN和HIF-1α蛋白在肺癌组织中表达及其临床意义。方法:应用免疫组化方法检测99例肺癌组织E-cd、PTEN和HIF-1α蛋白的表达情况。结果:E-cd、PTEN和HIF-1α蛋白在99例肺癌组织中的阳性率分别为63.6%(63/99)、62.6%(62/99)和56.6%(56/99),在癌旁正常组织中的阳性表达率分别为100%(20/20)、100%(20/20)和0,差异均有统计学意义,P<0.05。E-cd蛋白在细支气管肺泡癌组的表达高于小细胞癌组,而HIF-1α蛋白在细支气管肺泡癌组的表达低于小细胞癌组,差异均有统计学意义,P<0.05;其余各组织学类型之间3种蛋白表达差异均无统计学意义,P>0.05。E-cd和PTEN蛋白在淋巴结无转移组的表达均高于淋巴结转移组,P<0.05;而HIF-1α蛋白在两组间的表达差异无统计学意义,P>0.05。E-cd和PTEN蛋白在Ⅰ~Ⅱ期的表达高于Ⅲ~Ⅳ期,P<0.01;而HIF-1α蛋白在Ⅰ~Ⅱ期的表达低于Ⅲ~Ⅳ期,P<0.05。E-cd蛋白表达与PTEN蛋白表达呈正相关,P<0.05;HIF-1α蛋白与E-cd、PTEN蛋白表达均呈负相关,P<0.05。结论:E-cd、PTEN和HIF-1α蛋白的异常表达可能在肺癌的恶性演进中起重要作用,3者在肺癌的侵袭转移中可能起某种协同作用。
Objective: To investigate the expression of E-cadherin (E-cd), PTEN and HIF-1α in lung cancer and its clinical significance. Methods: The expressions of E-cd, PTEN and HIF-1α in 99 lung cancer tissues were detected by immunohistochemistry. Results: The positive rates of E-cd, PTEN and HIF-1α in 99 lung cancer tissues were 63.6% (63/99), 62.6% (62/99) and 56.6% (56/99), respectively. The positive expression rates in normal tissues were 100% (20/20), 100% (20/20) and 0 respectively, with statistical significance (P <0.05). The expression of E-cd protein in bronchioloalveolar carcinoma was higher than that in small cell carcinoma, while the expression of HIF-1α in bronchioloalveolar carcinoma was lower than that in small cell carcinoma (P <0.05). The remaining three histological types of three protein expression differences were not statistically significant, P> 0.05. The expression of E-cd and PTEN protein in lymph node metastasis group was higher than that in lymph node metastasis group, P <0.05; however, the expression of HIF-1α protein in the two groups had no significant difference (P> 0.05). The expression of E-cd and PTEN protein in stage Ⅰ ~ Ⅱ was higher than that in stage Ⅲ ~ Ⅳ, P <0.01; while the expression of HIF-1α in stage Ⅰ ~ Ⅱ was lower than that in stage Ⅲ ~ Ⅳ, P <0.05. E-cd protein expression was positively correlated with PTEN protein expression, P <0.05; HIF-1α protein was negatively correlated with E-cd, PTEN protein expression (P <0.05). Conclusion: The abnormal expression of E-cd, PTEN and HIF-1α may play an important role in the malignant progression of lung cancer. The three may play a synergistic role in the invasion and metastasis of lung cancer.
