目的:评估献血者血小板(PLT)采后计数实际测量值与AMICUS、TRIMA和MCS+血细胞分离机估算值之间的差异和符合性,以验证3种血细胞分离机采集血小板程序的安全性。方法:选取632名AMICUS献血者、575名TRIMA献血者和500名MCS+献血者分别在采后即刻、采后30min、采后1h留取1ml静脉血标本,动态观察单采后外周PLT的变化,从而比较机器的采后估算值与实测值之间的差异。结果:AMICUS、TRIMA和MCS+采后即刻,估算值与实测值正相差分别为4.86×109/L、4.95×109/L和4.97×109/L;采后30min,估算值与实测值负相差分别为2.51×109/L、2.35×109/L和2.57×109/L;采后1h,估算值与实测值负相差分别为7.54×109/L、7.44×109/L和7.52×109/L。采后即刻、采后30 min、采后1h,预测采后PLT的95%置信区间均大于100×109/L。结论:依照我国2011版新的“献血者健康检查要求”献血者采后PLT>100×109/L的采集标准,使用AMICUS、TRIMA和MCS 3种血细胞分离机采集PLT,将预测采后估算值作为安全捐献的指标,就目前的采集程序而言是安全可靠的。
PURPOSE: To assess the differences and compliance between actual measurements of PLT post-harvest counts and those estimated by the AMICUS, TRIMA and MCS + hemacytometer to verify the safety of platelet collection procedures by the 3 blood cell separators. Methods: 632 AMICUS blood donors, 575 TRIMA blood donors and 500 MCS + donors were enrolled in this study. The venous blood samples were collected immediately after the harvest, 30 min after the harvest and 1 mL after the harvest, and the peripheral PLT changes were observed dynamically. This compares the differences between the machine’s post-harvest estimates and the measured values. Results: The differences between the estimated and measured values were 4.86 × 109 / L, 4.95 × 109 / L and 4.97 × 109 / L for AMICUS, TRIMA and MCS + immediately after harvesting. The difference between the estimated and measured values was 2.51 × 109 / L, 2.35 × 109 / L and 2.57 × 109 / L, respectively. The negative correlation between the estimated value and the measured value was 7.54 × 109 / L, 7.44 × 109 / L and 7.52 × 109 / L, respectively, Immediately after harvest, 30 min after harvest and 1 h after harvest, the 95% confidence intervals of post-harvest PLT were all predicted to be greater than 100 × 109 / L. Conclusion: According to the collection standard of PLT> 100 × 109 / L after blood donors’ health checkup request in 2011, three kinds of blood cell separators were collected by AMICUS, TRIMA and MCS, Estimates, as an indicator of safe donation, are safe for the current collection procedure.
