目的研究高迁移率族蛋白N2(high mobility group chromosomal protein N2,HMGN2)对宫颈癌裸鼠移植瘤生长的抑制作用。方法建立宫颈癌裸鼠移植瘤模型,随机分为阴性对照组、HMGN2蛋白组、顺铂组和联合用药组,分别予生理盐水、HMGN2蛋白、顺铂、HMGN2蛋白加顺铂隔日腹腔注射。观察肿瘤的生长情况,经过4次给药后处死裸鼠,取出瘤块,测量其体积计算抑瘤率,并行苏木精-伊红(HE)染色观察形态学变化。结果成功建立宫颈癌裸鼠移植瘤模型。第4次给药后,HMGN2蛋白组、顺铂组、联合用药组肿瘤体积平均值分别为(0.14±0.07)cm3、(0.11±0.06)cm3、(0.11±0.07)cm3,与阴性对照组〔(0.38±0.12)cm3〕相比差异有统计学意义(P<0.05);3组肿瘤体积平均值的差异无统计学意义(P>0.05)。HMGN2蛋白组、顺铂组、联合用药组肿瘤抑制率分别为0.62±0.18、0.71±0.17、0.70±0.18,差异无统计学意义(P>0.05)。肿瘤经肉眼观察可见阴性对照组瘤块较大,HE染色可见HMGN2蛋白组、顺铂组和联合用药组细胞坏死较阴性对照组明显。结论 HMGN2蛋白可以明显抑制宫颈癌裸鼠移植瘤的生长。
Objective To investigate the inhibitory effect of high mobility group chromosomal protein N2 (HMGN2) on the growth of cervical cancer xenografts in nude mice. Methods The cervical cancer xenografts in nude mice were established and randomly divided into the negative control group, HMGN2 protein group, cisplatin group and combination group. The rats were injected intraperitoneally with normal saline, HMGN2 protein, cisplatin and HMGN2 plus cisplatin. The growth of the tumor was observed. After 4 times of administration, the nude mice were sacrificed, the tumor mass was removed, the tumor volume was measured, and the tumor inhibition rate was calculated. Morphological changes were observed by hematoxylin-eosin (HE) staining. Results Successful establishment of cervical cancer xenograft model in nude mice. After the fourth administration, the mean tumor volume of HMGN2 protein group, cisplatin group and combination group were (0.14 ± 0.07) cm3, (0.11 ± 0.06) cm3 and (0.11 ± 0.07) cm3, respectively. Compared with the negative control group (0.38 ± 0.12) cm3] (P <0.05). There was no significant difference in mean tumor volume between the three groups (P> 0.05). The tumor inhibition rates of HMGN2 protein group, cisplatin group and combination group were 0.62 ± 0.18,0.71 ± 0.17,0.70 ± 0.18, respectively, with no significant difference (P> 0.05). Negative control tumor tumor visible tumor mass larger, HE staining shows HMGN2 protein group, cisplatin group and combined treatment group necrosis than the negative control group. Conclusion HMGN2 protein can significantly inhibit the growth of cervical cancer xenografts in nude mice.
