骨髓基质细胞是骨髓造血微环境的重要组成成分。了解骨髓基质细胞的特性对进一步认识造血微环境调控造血的机理及其在病理条件下的变化具有重要意义。本文用单层液体培养方法培养小鼠骨髓基质细胞,并对其某些特性进行了初步观察。材料和方法动物:选用本院繁殖纯系LACA雄性小鼠,日龄60~70天。骨髓基质细胞培养体系:日本制R PMI1640双蒸水溶液8份,加1份经56℃灭活的小牛血清和1份马血清,外加青、链霉素各100微克/毫升,用碳酸氢钠溶液调整pH到7.2。
Bone marrow stromal cells are an important component of the hematopoietic microenvironment of the bone marrow. To understand the characteristics of bone marrow stromal cells to further understand the mechanism of hematopoietic microenvironment regulation of hematopoiesis and its pathological conditions of great significance. In this paper, single-layer liquid culture of mouse bone marrow stromal cells, and some of its characteristics were initially observed. MATERIALS AND METHODS Animal: LACA male mice were selected for propagation in our hospital and were aged 60-70 days. BMSCs culture system: 8 parts of R PMI1640 double distilled water solution made in Japan, 1 part of calf serum and 1 part of horse serum inactivated at 56 ℃, 100 μg / ml each of streptomycin and cyanine, The solution is adjusted to pH 7.2.
