目的研究人乳头状瘤病毒(HPV)16 E6 siRNA表达载体对宫颈癌Siha细胞株HPV16 E6基因的抑制作用。方法利用脂质体将HPV16E6特异性si RNA表达载体转染宫颈癌Siha细胞株,应用荧光定量RT-PCR检测HPV16 E6 mRNA的变化,流式细胞术检测E6蛋白表达的变化。结果细胞试验表明HPV16 E6siRNA表达载体可明显抑制Siha细胞E6 mRNA和E6蛋白的表达,抑制率分别为92.65%和84.40%。且其抑制效果可维持4周以上。结论HPV16 E6 siRNA表达载体可以高效、特异、长期的抑制宫颈癌Siha细胞HPV16 E6基因的表达。
Objective To study the inhibitory effect of human papillomavirus (HPV) 16 E6 siRNA on HPV16 E6 gene in cervical cancer Siha cell line. Methods HPV16E6 specific si RNA expression vector was transfected into Siha cells by lipofectamine. Fluorescent quantitative RT-PCR was used to detect the expression of HPV16 E6 mRNA. Flow cytometry was used to detect the expression of E6 protein. Results The results of cell assay showed that HPV16 E6 siRNA expression vector significantly inhibited the expression of E6 mRNA and E6 protein in Siha cells with the inhibitory rates of 92.65% and 84.40%, respectively. And its inhibitory effect can be maintained more than 4 weeks. Conclusion HPV16 E6 siRNA expression vector can efficiently, specifically and for a long time inhibit the expression of HPV16 E6 gene in Siha cells.
