目的寻找小桐子Jatropha curcas果壳的抑菌活性物质。方法利用6种不同极性溶剂对果壳进行平行提取,结合抑菌活性测定,筛选出活性最强、提取率大于10%的粗提物;再将筛选出的粗提物,依次利用液-液萃取、硅胶柱反复柱色谱,结合PTLC制备分离纯化。同时对各阶段分离产物进行活性追踪,筛选出具有抗菌活性的物质,最后采用HPLC-MS分析,初步鉴定出活性部位主体成分。结果从果壳中分得抑菌活性强且含量高WA3物质,经HPLC-MS分析,初步鉴定了含量较高的3个化合物,为总量的35.27%,分别为环庚肽、麻疯树素、三萜醇三聚体。结论小桐子果壳中含有丰富的抑菌活性物质,分别属于肽类、二萜、三萜类化合物。
Objective To search for antibacterial substances of Jatropha curcas husk. Methods Using six kinds of solvents with different polarity to extract the shells in parallel, combined with the determination of antibacterial activity, the crude extracts with the strongest activity and the extraction rate of more than 10% were screened out; Liquid extraction, silica gel column chromatography, separation and purification in combination with PTLC preparation. At the same time, the activity of the separated products at each stage was followed, and the substances with antibacterial activity were screened. Finally, the main components of the active site were identified by HPLC-MS analysis. RESULTS: From the fruit shell, strong antibacterial activity and high content of WA3 were obtained. According to HPLC-MS analysis, 3 compounds with higher content were identified, which was 35.27% of the total, which were cycloheptapeptide and jatropha curcas. Prime, triterpene alcohol trimer. Conclusion Jatropha curcas contains rich antibacterial active substances, belonging to peptides, diterpenoids and triterpenoids.
