目的:探讨激活大麻受体对肝癌细胞增殖和凋亡的作用,并对其机制进行初步探讨。方法:将细胞分成对照组、不同剂量大麻受体激动剂WIN-55,212-2(WIN)处理组。MTT法测定WIN对HepG2细胞增殖的影响;DNA梯度电泳法检测HepG2细胞凋亡DNA片段,流式细胞术检测分析各组细胞凋亡率变化,Western blot分析各组细胞caspase-3和c-myc的蛋白表达,分光光度法检测caspase-3、8、9的活性。结果:大麻受体激动剂WIN能抑制肝癌细胞生长和c-myc的蛋白表达,诱导肝癌细胞凋亡,上述效应具有剂量依赖性。而且WIN可能通过诱导细胞caspase-3的蛋白表达和激活caspase-3、8、9活性进而诱导肝癌细胞凋亡。结论:大麻受体激动剂WIN能抑制肝癌细胞生长,诱导肝癌细胞凋亡,并与caspases和c-myc的蛋白表达相关。
Objective: To investigate the effect of activating marijuana receptor on the proliferation and apoptosis of hepatocellular carcinoma cells, and to explore its mechanism. Methods: The cells were divided into control group and different doses of cannabinoid receptor agonist WIN-55,212-2 (WIN). MTT method was used to determine the effect of WIN on the proliferation of HepG2 cells; DNA gradient electrophoresis was used to detect the apoptotic DNA fragments of HepG2 cells; flow cytometry was used to detect the changes of apoptosis rate; Western blot was used to analyze the expressions of caspase-3 and c-myc Of protein expression, spectrophotometry assay caspase-3,8,9 activity. Results: Cannabinoid receptor agonist WIN could inhibit the growth of hepatocellular carcinoma cells and the expression of c-myc protein, and induce the apoptosis of hepatocellular carcinoma cells in a dose-dependent manner. Moreover, WIN may induce hepatocellular carcinoma cell apoptosis by inducing caspase-3 protein expression and activating caspase-3, 8 and 9 activity. Conclusion: Cannabinoid receptor agonist WIN can inhibit the growth of hepatoma cells and induce the apoptosis of hepatocellular carcinoma cells, and is related to the protein expression of caspases and c-myc.
