目的观察二烯丙基二硫(DADS)对乳腺癌BT474细胞生长、侵袭的影响及其机制。方法培养人乳腺癌BT474细胞,采用逆转录定量聚合酶链反应(qRT-PCR)法检测不同浓度DADS处理的BT474细胞中的GABARAPL1表达变化,采用甲基噻唑基四唑(MTT)法检测各组细胞增殖情况。结果 50、100、200和400μmol/L浓度DADS处理的BT474细胞GABARAPL1相对表达量低于0μmol/L组,差异有统计学意义(P<0.05)。GABARAPL1干扰组、DADS组、GABARAPL1干扰组+DADS组BT474细胞OD570值低于siRNA阴性对照组,差异有统计学意义(P<0.05)。结论 DADS通过下调GABARAPL1的表达抑制乳腺癌BT474细胞的增殖。
Objective To observe the effect of diallyl disulfide (DADS) on the growth and invasion of breast cancer BT474 cells and its mechanism. Methods Human breast cancer BT474 cells were cultured and the expression of GABARAPL1 in BT474 cells treated with different concentrations of DADS was detected by reverse transcription quantitative polymerase chain reaction (qRT-PCR). MTT assay was used to detect the expression of GABARAPL1 Cell proliferation. Results The relative expression levels of GABARAPL1 in BT474 cells treated with DADS at 50, 100, 200 and 400 μmol / L were lower than those at 0 μmol / L (P <0.05). The OD570 value of BT474 cells in GABARAPL1 interference group, DADS group, GABARAPL1 interference group and DADS group was lower than that in siRNA negative control group, the difference was statistically significant (P <0.05). Conclusion DADS inhibits the proliferation of breast cancer BT474 cells by down-regulating GABARAPL1 expression.
