目的:制备乙酰化苦豆子多糖,并对修饰后的乙酰化苦豆子多糖溶液的构象特征进行研究。方法:对苦豆子多糖进行乙酰化修饰,采用取代度测定、红外光谱分析手段对修饰结果进行验证,采用碘-碘化钾反应和刚果红试验研究乙酰化苦豆子多糖的构象特征,采用圆二色谱法(CD)对不同因素影响下乙酰化苦豆子多糖的溶液行为进行研究。结果:乙酰化苦豆子多糖修饰成功,取代度为0.84,其溶液行为显示乙酰化苦豆子多糖具有多股螺旋结构以及较长的侧链和较多的分支,并且溶液构象受浓度、温度以及添加金属离子的影响而变化。结论:成功修饰获得乙酰化苦豆子多糖,并对其溶液构象进行了全面研究,为其进一步研究提供了参考。
OBJECTIVE: To prepare acetylated Sophora alopecuroides polysaccharide and to study the conformational characteristics of the modified acetylated Sophora alopecuroides solution. Methods: The acetylated polysaccharides of Sophora alopecuroides were modified by the methods of degree of substitution and infrared spectroscopy. The conformational characteristics of acetylated Sophora alopecuroides polysaccharide were studied by iodine-potassium iodide reaction and Congo red test. (CD) on the behavior of acetylated Sophora alopecuroides polysaccharide under different factors. Results: Acetylated Sophora alopecuroid polysaccharide was successfully modified with a degree of substitution of 0.84. The solution behavior of acetylated Sophora alopecuroid polysaccharide showed multi-stranded helical structure with longer side chains and more branches. The solution conformation was affected by concentration, temperature and addition Metal ions vary. Conclusion: The acetylated Sophora flavescens polysaccharides were successfully modified and their solution conformations were studied comprehensively, which provided a reference for further research.
