目的研究颐脑解郁方对抑郁模型大鼠脑内胶质原纤维酸性蛋白(GFAP)、碱性成纤维细胞生长因子(bFGF)及脑源性神经营养因子(BDNF)表达的影响。方法二级雄性Wistar大鼠分为正常组、抑郁组、治疗组、对照组。抑郁组、治疗组、对照组采用慢性应激方法结合孤养法造成抑郁大鼠模型,治疗组每日灌服中药颐脑解郁方1次,连续给药6周,对照组每日灌服生理盐水1次,连续6周。取脑组织,采用免疫组化方法,测定各组大鼠脑额叶皮质及海马GFAP、bFGF、BDNF的表达。结果对照组大鼠脑额叶皮质及海马GFAP、bFGF、BDNF阳性细胞数量减少,表达减弱,细胞大小不均,排列不规则。治疗组大鼠脑额叶皮质及海马GFAP、bFGF、BDNF阳性细胞数目显著增加,形态规则,表达增强;对GFAP、bFGF、BDNF阳性细胞进行测量分析,统计每个场的面密度,与对照组相比,有统计学意义(P<0.05或P<0.01)。结论颐脑解郁方通过增强模型大鼠脑内GFAP、bFGF、BDNF的表达,促进5-羟色胺、去甲肾上腺素的功能,从而改善抑郁症状。
Objective To investigate the effects of Yinaojieyu Formula on the expression of glial fibrillary acidic protein (GFAP), basic fibroblast growth factor (bFGF) and brain-derived neurotrophic factor (BDNF) in the rat model of depression. Methods Two male Wistar rats were divided into normal group, depression group, treatment group and control group. The depression group, the treatment group and the control group were induced by the chronic stress method and the lone solitary method. The treatment group were given the traditional Chinese medicine Yinao Jieyu Decoction once daily for 6 weeks, while the control group were fed with normal saline 1 times for 6 weeks. The brain tissue was taken out and the expression of GFAP, bFGF and BDNF in the cortex and hippocampus were determined by immunohistochemistry. Results The number of GFAP, bFGF and BDNF positive cells in the frontal cortex and the hippocampus in the control group decreased, the expressions were weakened, the cells were irregular in size and arranged irregularly. The number of GFAP, bFGF and BDNF positive cells in the frontal cortex and hippocampus of rats in treatment group were significantly increased and the morphology was enhanced. The GFAP, bFGF and BDNF positive cells were measured and analyzed. The surface density of each field was calculated and compared with the control group Compared with the statistical significance (P <0.05 or P <0.01). Conclusion Yinao Jieyu decoction can improve depressive symptom by enhancing the expression of GFAP, bFGF and BDNF in brain of model rats, promoting the function of serotonin and norepinephrine.
