目的研究在非小细胞肺癌(non-small cell lung cancer,NSCLC)中,Axin(Axis inhibitor)与细胞凋亡的关系以及X射线能否上调NSCLC组织中Axin表达,并促进NSCLC细胞凋亡。方法用免疫组织化学和TUNEL(TdT-mediated dUTP-X射nick end labeling)法检测95例NSCLC组织内Axin的表达及凋亡情况。将新鲜的NSCLC组织及培养的细胞系(A549和BE1)用X射线照射后,用免疫印迹和RT-PCR方法检测组织中Axin、凋亡蛋白caspase-3表达,并用流式细胞仪检测细胞凋亡情况。结果在hNSCLC组织内,可见少量凋亡细胞和Axin阳性细胞,凋亡与Axin的表达呈正相关。X射线照射后,部分病例的hNSCLC组织内Axin表达明显上调并促进A549和BE1细胞系凋亡。结论 X射线能诱导NSCLC组织Axin表达上调并促进A549和BE1细胞凋亡。
Objective To investigate the relationship between Axin (Axis inhibitor) and apoptosis in non-small cell lung cancer (NSCLC) and whether X-ray can up-regulate Axin expression in NSCLC and promote apoptosis of NSCLC. Methods The expression of Axin and apoptosis in 95 NSCLC tissues were detected by immunohistochemistry (TUNEL) and nick end labeling (TUNEL). The fresh NSCLC tissues and cultured cell lines (A549 and BE1) were irradiated with X-rays. Axin and caspase-3 protein expression in the tissue were detected by Western blotting and RT-PCR. Flow cytometry was used to detect the apoptosis Death situation. Results A small amount of apoptotic cells and Axin positive cells were found in hNSCLC tissues. Apoptosis was positively correlated with Axin expression. After X-ray irradiation, Axin expression was significantly upregulated in hNSCLC tissues in some cases and apoptosis of A549 and BE1 cell lines was promoted. Conclusion X-ray can induce Axin expression in NSCLC and promote the apoptosis of A549 and BE1 cells.
