目的观察巴戟天含药血清对原代破骨细胞RANK和CA Ⅱ mRNA表达的影响。方法取SPF级大鼠48只,随机分成正常组12只和去势组36只,正常组切去卵巢周围部分脂肪,去势组切除卵巢。3个月后测定两组雌激素水平并将去势组随机分为骨质疏松组,骨质疏松+雌激素组,骨质疏松+巴戟天含药血清组。采用机械分离法提取各组的破骨细胞,培养5 d后,TRAP染色及电镜扫描骨片等的方法鉴定破骨细胞。最后用雌激素或巴戟天含药血清干预3 d,以RT-PCR法检测各组RANK和CA ⅡmRNA表达。采用单因素方差分析或多样本均数两两比较进行统计分析。结果去势组后大鼠雌激素水平低于正常组(P<0.01)。骨质疏松组破骨细胞RANK和CA Ⅱ表达均高于正常组(P<0.05,P<0.05);巴戟天含药血清可降低骨质疏松后大鼠破骨细胞RANK和CA II的表达(P<0.05,P<0.05)。结论巴戟天和雌激素均可降低骨质疏松大鼠破骨细胞RANK和CA Ⅱ的表达,从而达到抑制骨质疏松的作用。
Objective To observe the effect of medicinal serum of Morinda officinalis on the expression of RANK and CA Ⅱ mRNA of primary osteoclasts. Methods Forty-eight SPF rats were randomly divided into normal control group (n = 12) and castration group (n = 36). Normal group was given partial fat removal and ovariectomized group. Three months later, the levels of estrogen in two groups were determined and the castration group was randomly divided into osteoporosis group, osteoporosis + estrogen group and osteoporosis + Morinda officinalis serum group. The osteoclasts of each group were extracted by mechanical separation, and cultured for 5 days. TRAP staining and scanning electron microscope were used to identify osteoclasts. Finally, with estrogen or Morinda officinalis serum intervention 3 d, RT-PCR method to detect the expression of RANK and CA Ⅱ mRNA in each group. One-way ANOVA or multiple comparisons were used for statistical analysis. Results The level of estrogen in the castration group was lower than that in the normal group (P <0.01). The expressions of RANK and CA Ⅱ in osteoclasts of osteoporosis group were higher than those in normal group (P <0.05, P <0.05). The medicinal serum containing Morinda officinalis decreased the expression of RANK and CA II in osteoclasts of osteoporosis rats (P <0.05, P <0.05). Conclusion Both Morinda officinalis and estrogen can reduce the expression of RANK and CA Ⅱ in osteoclasts of rats with osteoporosis, so as to inhibit the osteoporosis.
