为建立裂叶榆组织培养体系,选取裂叶榆木质化和水培萌发幼嫩茎段进行离体培养,探讨不同消毒时间、激素种类和浓度对裂叶榆离体快繁的影响。表明:木质化茎段在0.1%升汞消毒14 min时,污染率稍低,褐化率低,成活率高;幼嫩茎段在0.1%升汞消毒3 min时幼嫩茎段污染率最低,褐化率较低;在对休眠芽和幼嫩茎段进行离体培养时筛选出MS+2.0 mg/L 6-BA+0.1 mg/L NAA为最佳培养基激素组合,且在BA:NAA浓度比为20:1时,培养15 d后,裂叶榆的腋芽萌发率最高(80%),而且萌发芽的腋芽绿色,长势良好。
In order to establish the tissue culture system of Crataegus tenuipes, the young stem segments of Ulmus pumila was selected for in vitro culture, and the effects of different disinfection time, hormone types and concentrations on the rapid propagation of Ulmus pumilas were studied.
