目的建立高效液相色谱法(HPLC)同时测定无梗五加中7个有机酸(绿原酸,新绿原酸,咖啡酸,1,3-二咖啡酰奎宁酸,异绿原酸B,异绿原酸A,异绿原酸C)含量的方法。方法采用Hypersil ODS_2 C_(18)(250 mm×4.6 mm,5μm)色谱柱,柱温室温,以O.1%甲酸乙腈-O.1%甲酸水溶液进行梯度洗脱,流速1.0 mL/min,检测波长324 nm。结果 7个有机酸的线性范围分别为20.5~328、1.99~31.8、2.43~38.9、2.44~39.0、2.52~40.3、7.25~116、2.42~38.7 mg/L,平均加样回收率均在95.2%~103.9%范围内。结论该方法快速简便、精密度好、灵敏度高,为控制无梗五加药材质量及与五加属的其他药材进行对比研究提供了方法。
OBJECTIVE To establish a HPLC method for the simultaneous determination of 7 organic acids (chlorogenic acid, neogenic acid, caffeic acid, 1,3-dicaffeoylquinic acid, iso-chlorogenic acid B, Iso-chlorogenic acid A, iso-chlorogenic acid C) content of the method. Methods A Hypersil ODS_2 C_ (18) (250 mm × 4.6 mm, 5 μm) column was used. The column was eluted with 0.1% formic acid in acetonitrile and 0.1% formic acid at a flow rate of 1.0 mL / Wavelength 324 nm. Results The linear range of seven organic acids were 20.5 ~ 328, 1.99 ~ 31.8, 2.43 ~ 38.9, 2.44 ~ 39.0, 2.52 ~ 40.3, 7.25 ~ 116 and 2.42 ~ 38.7 mg / L, ~ 103.9% range. Conclusion The method is rapid, simple, accurate and sensitive. It provides a method for controlling the quality of medicinal materials without sequela and comparing with other medicinal materials of Genus.
