本实验应用放射免疫方法,对雄性成年SD大鼠离体下丘脑、腺垂体、睾丸三级组织进行培养顺序灌注,观察P物质对于促性腺激素释放激素(GnRH)、黄体生成素(LH)及睾酮(T)分泌的影响。将实验动物50只,随机分成五组。前三组进行下丘脑、腺垂体培养;后两组进行腺垂体、睾丸培养。将动物断头处死,在超静工作台上立即取出下丘脑组织(包括视前区和下丘脑中央基底部)腺垂体组织及一侧睾丸组织。分别装入各自对应的预盛有5ml新鲜完全1640液的培养板孔中,向其中第二组下丘脑培养液中加入1ngSP后,进行培养。条件:95%O2、5%CO2;温度:37C;pH:7.2—7.4,3小时后,将各组下一级培养液取出2ml待测。同时从上一级培养液中取出1ml与1ml新鲜完全1640液共同加入到下一级培养液中,并对第三、五组同时向下一级培养液加入1ngSP,再培养3小时后,取出各自的2ml培养液进行放免测定,与上一级取出液的测量值进行比较。实验结果:第一组LH含量增值为(46.08+3.91)mIU/ml,第二组(64.29+4.77)mIU/ml,第三组(29.50+3.13)mIU/ml,第四组T含量增量为(3.94+0.52)ng/ml?
In this experiment, we used radioimmunoassay to perfuse the hypothalamus, adenohypophysis and testis in male adult SD rats in order to observe the effects of substance P on the expression of GnRH, LH, Effects of Testosterone (T) Secretion. 50 experimental animals were randomly divided into five groups.
