目的 探讨转化生长因子 β1(TGF β1)对人胚腱细胞增殖的调控作用。方法 采用3H TdR掺入法观察TGF β1对人胚腱细胞的DNA合成剂量效应和时间效应 ;采用流式细胞仪观察TGF β1对细胞周期及增殖指数PI值 (S +G2M)的影响。结果 在 0 .5 %胎牛血清条件下TGF β1可刺激腱细胞增殖 ,其作用在 0 .5~ 10ng/ml之间呈剂量依赖性增强 ,当剂量为 10ng/ml时 ,促DNA合成作用达到饱和。 10ng/mlTGF β1作用 36h时开始增殖显著 ,并可持续到 72h。细胞周期分析表明 :在TGF β1作用下 ,G0 /G1%呈降低趋势 ,而S %呈升高趋势 ,反映细胞增殖活力的PI值也呈升高趋势 ,尤以 5ng/ml以上浓度时为显著。结论 TGF β1可能通过促进腱细胞增殖参与肌腱的修复
Objective To investigate the regulatory effect of transforming growth factor-β1 (TGF-β1) on the proliferation of human embryo tenocytes. Methods DNA synthesis dose-effect and time-effect of TGF-β1 on human embryo tendon cells were observed by 3H TdR incorporation method. The effect of TGF-β1 on the cell cycle and proliferation index PI (S + G2M) was observed by flow cytometry. Results TGFβ1 stimulated the proliferation of tendon cells at 0. 5% fetal bovine serum, and its effect increased from 0 to 10ng / ml in a dose-dependent manner. When the dosage was 10ng / ml, the effect of DNA synthesis was promoted saturation. The proliferation of TGF-β1 at 10ng / ml began to proliferate significantly at 36h, and continued to 72h. Cell cycle analysis showed that under the action of TGFβ1, the G0 / G1% showed a decreasing trend while the S% showed a rising trend, while the PI value reflecting the cell proliferation activity also tended to increase, especially at a concentration above 5ng / ml . Conclusion TGF β1 may participate in the repair of tendon by promoting the proliferation of tendon cells
