本实验研究了一种体外制备人工关节金属磨损颗粒的方法,验证其在人工关节无菌性松动的实验研究中应用的可行性.[方法]通过真窄球磨法体外制备人工关节金属磨损颗粒,同时对磨损颗粒进行元素分析,扫描电镜观察,激光粒度仪检测,并与RAW264.7细胞共培养,检测细胞毒性.[结果]真空球磨法体外制备人工关节金属磨损颗粒直径<5μm,与体内分离的人工关节金属磨损颗粒理化性质一致,在试验浓度(通常低于1mg/ml)没有显示细胞毒性.[结论]真空球磨法体外制备人工关节金属磨损颗粒简便易行,重复性好,可以用于相关的细胞学研究.“,”[Objective]To investigate a method of in vitro preparation and separation for metallic wear particles around joint prosthesis,and evaluate its feasibility in medical experiments of prosthetic loosening.[Method]Metallic wear particles were made around joint prosthesis by vacuum ball mill in vitro.Particle size,size distribution,chemical composition and shape were charac-terized by element trace analysis,laser eountersizer,scanning electron microscopy.Metal particles were co-cultured with RAW264.7 cells,and its eytotoxieity was detected.[Result]Metal particles made by vacuum ball mill in vitro was similar to par-tieles extracted in vivo(<5μm in diameter).No cell eytotoxieity Wag found in experimental concentration(<1.0mg/ml).[Conclusion]Metallic wear particles made in vitro is similar to that in vivo,it can be used in vivo studies of cytology.
