目的:建立较为全面反映肝星状细胞(hepatic stellate cell,HSC)活化效应的细胞模型。方法:应用脂多糖(lipopolysccharide,LPS)刺激体外培养的大鼠HSC。噻唑蓝(MTT)法测定HSC增殖率。免疫组化法显示HSC表达其Ⅰ、Ⅲ型胶原的表达水平,并进行图象分析。结果:LPS明显刺激HSC的增殖并提高Ⅰ、Ⅲ型胶原表达水平,与对照组比较差异有非常显著性意义(P<0.01)。结论:LPS诱导HSC活化模型能够相对全面地反映HSC活化的效应。
Objective: To establish a cell model that reflects the activation of hepatic stellate cells (HSC). Methods: Rat HSC cultured in vitro was stimulated with lipopolysaccharide (LPS). The proliferation rate of HSC was determined by MTT assay. Immunohistochemistry showed that HSC expressed its type Ⅰ and type Ⅲ collagen expression and image analysis. Results: LPS significantly stimulated the proliferation of HSC and increased the expression of type Ⅰ and type Ⅲ collagen, which was significantly different from the control group (P <0.01). Conclusion: LPS-induced HSC activation model can reflect the effect of HSC activation relatively completely.
