目的研究淫羊藿、女贞子协同布地奈德对哮喘大鼠糖皮质激素受体(GR)及抗炎作用的影响。方法 SPF级健康雄性SD大鼠,随机分为正常对照组、哮喘模型组、布地奈德组(0.01%布地奈德雾化20 min)、煎剂组(淫羊藿、女贞子煎剂3.5 g/kg灌胃)、布地奈德合煎剂组(0.01%布地奈德雾化20 min+淫羊藿、女贞子煎剂3.5 g/kg灌胃)。采用酶免法检测血清皮质酮(CORT)、白介素-4(IL-4)、免疫球蛋白E(Ig E)含量,流式细胞术检测肺泡灌洗液(BALF)中GR蛋白表达及结合力。结果布地奈德组及布地奈德合煎剂组可显著降低血清IL-4、Ig E含量(P<0.05,P<0.01);煎剂组能显著降低血清Ig E含量(P<0.05)、升高血清CORT、BALF GR蛋白含量(P<0.05);布地奈德合煎剂组能显著升高血清CORT含量、BALF GR蛋白含量及蛋白结合力(P<0.01,P<0.05)。结论保护糖皮质激素(GC)发挥作用的环节及局部抗炎作用的敏感性可能是淫羊藿、女贞子协同布地奈德治疗哮喘的作用机制之一。 Objective To study the effects of Epimedium, Fructus Ligustri Lucidi and budesonide on glucocorticoid receptor (GR) and anti-inflammatory effects in asthmatic rats. Methods SPF healthy male Sprague-Dawley rats were randomly divided into normal control group, asthma model group, budesonide group (0.01% budesonide atomization 20 min), decoction group (Epimedium, Ligustrum lucidum decoction 3.5 g / kg orally), budesonide combined decoction group (0.01% budesonide atomization 20 min + Epimedium, Ligustrum lucidum decoction 3.5 g / kg gavage). Serum corticosterone (CORT), interleukin-4 (IL-4) and IgE were detected by enzyme-linked immunosorbent assay. GR protein expression and binding capacity in BALF were detected by flow cytometry . Results The budesonide group and budesonide combined with decoction group could significantly reduce the levels of serum IL-4 and Ig E (P <0.05, P <0.01), decoction group could significantly reduce the serum IgE level (P <0.05) (P <0.05). The budesonide decoction group could significantly increase the serum CORT content, the content of BALF GR protein and the protein binding ability (P <0.01, P <0.05). Conclusion The protective effect of glucocorticoid (GC) and the sensitivity of local anti-inflammatory effect may be one of the mechanisms of Epimedium, Ligustrum lucidum and budesonide in the treatment of asthma.
