建立了紫外-可见光分光光度法测定去铁胺含量的方法。去铁胺在HAc-NAc缓冲溶液中,与Fe(II)发生显色反应,采用紫外-可见光分光光度法测定其含量。结果表明,去铁胺在0.0897~0.8074 mg/m L范围内,呈现良好的线性关系,回归方程为Y=9.5714X-0.0069(R2=0.9988)。在供试液中分别添加去铁胺对照品0.1,0.2,0.3 mg/m L 3个水平添加下,平均回收率99.5%,相对标准偏差(RSD)为2.0%。方法可用于甲磺酸去铁胺制剂中去铁胺的质量控制。
A method for the determination of deferoxamine by ultraviolet-visible spectrophotometry was established. Deferoxamine in HAc-NAc buffer solution, with Fe (II) color reaction, the determination of its content by UV-Vis spectrophotometry. The results showed that the deferoxamine showed a good linearity in the range of 0.0897 ~ 0.8074 mg / mL, the regression equation was Y = 9.5714X-0.0069 (R2 = 0.9988). The average recoveries were 99.5% and the relative standard deviations (RSDs) were 2.0% with addition of deferoxamine 0.1, 0.2 and 0.3 mg / mL respectively. The method can be used for quality control of deferoxamine in deferoxamine mesylate preparations.
