目的 研究阿司匹林对TNFα 诱导的内皮细胞与中性粒细胞及单核细胞粘附的抑制作用及作用机制。方法 用髓过氧化酶法测定白细胞与内皮细胞的粘附 ,ELISA法测定内皮细胞粘附分子的表达。结果 阿司匹林(6 0 0~ 90 0mg·L-1)可抑制中性粒细胞及单核细胞与TNFα激活 6h的内皮细胞粘附 ;而TNFα 激活内皮细胞后 2 4h ,阿司匹林对单核细胞与其粘附的抑制作用增强但却不抑制中性粒细胞与其粘附。结论 阿司匹林通过抑制E selectin的表达而降低TNFα诱导的内皮细胞与中性粒细胞的粘附 ,并通过抑制VCAM 1的表达降低TNFα诱导的内皮细胞与单核细胞的粘附。
Objective To study the inhibitory effect and mechanism of aspirin on TNFα-induced endothelial cell adhesion to neutrophils and monocytes. Methods The myeloperoxidase method was used to determine the adhesion of leukocytes to endothelial cells. The expression of endothelial cell adhesion molecules was measured by ELISA. Results Aspirin (6 0 0 ~ 90 0 mg · L -1) inhibited the adhesion of neutrophils and monocytes to TNFα for 6 h, while the effect of aspirin on mononuclear cells and its adhesion 24 h after TNFα activation The attached inhibitory effect is enhanced but does not inhibit neutrophil adhesion. Conclusion Aspirin can reduce the TNFα-induced adhesion of endothelial cells to neutrophils by inhibiting the expression of E selectin, and decrease the TNFα-induced adhesion of endothelial cells to monocytes by inhibiting the expression of VCAM 1.
