目的用HPLC法同时测定独活寄生丸中的龙胆苦苷和芍药苷。方法采用C18色谱柱,以乙腈-0.1%磷酸(12∶88)为流动相,流速1.5 mL·min-1,检测波长0~15 min为270 nm(龙胆苦苷),15.01~30 min为230 nm(芍药苷)。结果样品中龙胆苦苷和芍药苷能够达到基线分离,进样量分别为0.1643~3.2860μg、0.0642~1.2830μg时与峰面积呈良好的线性关系(r=0.9999),平均回收率分别为97.2%、97.8%,RSD分别为1.7%、1.9%。结论所用方法快速、简便,重复性好,可用于独活寄生丸的质量控制。
OBJECTIVE To determine gentiopicroside and paeoniflorin in Duhuo parasitic pills simultaneously by HPLC method. Methods The mobile phase was acetonitrile-0.1% phosphoric acid (12:88) at a flow rate of 1.5 mL · min-1 on a C18 column with a detection wavelength of 270 nm (gentiopicroside) at 0-15 min. The detection wavelength was 15.01-30 min 230 nm (paeoniflorin). Results The gentiopicroside and paeoniflorin in the sample were able to achieve the baseline separation. The injection volumes were 0.1643 ~ 3.2860μg and 0.0642 ~ 1.2830μg, respectively, and the peak area was linear (r = 0.9999). The average recoveries were 97.2 %, 97.8%, RSD 1.7%, 1.9% respectively. Conclusion The method is rapid, simple and reproducible, and can be used for the quality control of live alone parasitic pills.
