目的测定和比较不同正常细胞及人类肿瘤细胞膜上尿型纤溶酶原活化素受体(uPAR)的表达,了解肿瘤细胞表达uPAR的基本性质。方法建立并应用受体放射配基结合分析法(receptor- radio- ligand binding assay, RBA)测定细胞膜上的uPAR。结果 成纤维细胞经不同处理,U937细胞以PMA处理不同时间,测定uPAR的结果无论表达量或平衡解离常数(Kd)的变化均与文献一致。对正常细胞和肿瘤细胞株共10种测定膜上uPAR,结果肺癌、肝癌等5种人类肿瘤细胞比人成纤维细胞、牛内皮细胞等正常组织细胞的表达数明显多。结论 本研究进一步证明了uPAR表达增强是多种恶性肿瘤细胞的一种特点。
Objective To determine and compare the expression of urinary plasminogen activator receptor (uPAR) in different normal human and human tumor cell membranes and to understand the basic properties of uPAR expression in tumor cells. Methods To establish and apply receptor-radio-ligand binding assay (RBA) to determine the uPAR on the cell membrane. Results After treated with different concentrations of fibroblasts, U937 cells were treated with PMA at different times. The results of determination of uPAR were consistent with those in the literature, regardless of the expression or the equilibrium dissociation constant (Kd). A total of 10 normal and tumor cell lines were assayed for uPAR, resulting in significantly higher expression of five human tumor cell lines, lung cancer and liver cancer, than normal human cells, such as human fibroblasts and bovine endothelial cells. Conclusions This study further demonstrates that enhanced uPAR expression is a hallmark of many malignant tumor cells.
