【目的】针对两个典型的水稻品种(系)耐盐材料(02-11和津源85)和两个敏感材料(秋田小町和99-28),进行水稻耐盐相关的SSR标记的研究。【方法】选取水稻12条染色体上的565对SSR引物进行亲本间多态性筛选;选择适合的标记引物,并确定PCR扩增的最适退火温度。【结果】在565对引物组合中筛选出68对引物在4个材料间有多态性;明确了主要标记引物的通用退火温度为55℃。【结论】68对可作为该水稻群体耐盐QTL分析的SSR标记引物,为进一步进行水稻耐盐性分子鉴定和耐盐分子辅助育种提供参考。
【Objective】 Two salt-tolerant materials (02-11 and Jinyuan 85) and two sensitive materials (Koshima Akita and 99-28) were used to study salt tolerance-related SSR markers in rice. 【Method】 565 pairs of SSR primers on 12 chromosomes of rice were selected for polymorphism screening. The suitable primers were selected and the optimum annealing temperature for PCR amplification was determined. 【Result】 Sixty-six pairs of primer combinations were screened out of 565 pairs of primers with polymorphism among the four materials. The common annealing temperature of the major labeled primers was 55 ℃. 【Conclusion】 Sixty-eight SSR markers could be used as SSR markers for salt-tolerance QTL analysis in this rice population, providing a reference for further molecular identification of salt tolerance and salt-tolerant molecular breeding in rice.
