目的分析中华(江苏)骨髓库HLAⅠ类抗原的分布特点。方法采用序列特异性引物(PCR SSP)基因分型技术,对中华(江苏)骨髓库初期的骨髓捐献志愿者的HLA A、B位点上的等位基因作低分辨基因分型,计算HLA A、B的基因频率、由基因分型结果指定的抗原特异性频率(以下简称抗原频率)、HLA A、B位点单倍型频率和连锁不平衡参数,获取中华(江苏)骨髓库HLAⅠ类抗原多态性分布的初步资料。结果中华(江苏)骨髓库志愿者中,A位点抗原频率较高的是A2、A11和A24,分别为0.5087,0.3505和0.3056;而A74的频率最低(0.0009);B位点抗原频率较高的是B13、B46和B60,分别为0.2372,0.1772和0.1603,频率较低的是B78(0.0002)。连锁不平衡参数>0的单倍型有80余种,最常见的单倍型有A33B58、A2B62、A2B13、A33B44、A30B13等。结论了解中华(江苏)骨髓库志愿者HLA遗传学分布特点,为器官或干细胞移植配型和疾病相关性分析提供了有价值的基础性资料。
Objective To analyze the distribution of HLA class I antigens in China (Jiangsu) bone marrow library. Methods The alleles of HLA A and B loci in the bone marrow donor volunteers of China (Jiangsu) bone marrow bank were analyzed by PCR SSP genotyping. HLA A , B gene frequency, the specific antigen-specific frequency (hereinafter referred to as the antigen frequency), HLA A, B haplotype frequencies and linkage disequilibrium parameters, to obtain China (Jiangsu) bone marrow HLA class I antigen Polymorphism distribution of the preliminary data. Results Among the bone marrow pool volunteers in China (Jiangsu), the frequency of allele A was A2, A11 and A24 were 0.5087, 0.3550 and 0.3056 respectively, while the frequency of A74 was the lowest (0.0009). The frequency of B antigen was higher B13, B46 and B60 were 0.2372, 0.1772 and 0.1603, respectively. The lower frequency was B78 (0.0002). There are more than 80 haplotypes with linkage disequilibrium parameter> 0. The most common haplotypes are A33B58, A2B62, A2B13, A33B44, A30B13 and so on. Conclusion The HLA genotypic distribution of Chinese (Jiangsu) Marrow Strain Volunteers is helpful to provide valuable basic information for the analysis of the matching of organ and stem cell transplantation and disease.
